TCX-201 and TCX-101 data presented at AACR 2026

Poster 5631 — TCX-201
TCX-201, a novel ADC targeting a specific tumor-associated carbohydrate antigen (TACA) with excellent preclinical efficacy, PK and safety profile in solid tumor models.

- TCX-201 was tested with three payloads (MMAE DAR4, exatecan DAR8, deruxtecan DAR8). In vitro potency tracked the amount of target on the cell surface, including patient-derived cell lines from colorectal (2 of 5) and pancreatic (6 of 8) tumors.
- The MMAE variant reduced tumor growth dose-dependently in cell line-derived xenografts (T/C of 5.7 % at 7 mg/kg in Capan-2 and 2.8 % in Colo-205, with 33 % complete responses in Colo-205).
- In a patient-derived xenograft from metastatic colorectal cancer, TCX-201 MMAE reached a complete response rate above 80 % at 5 and 10 mg/kg.
- Half-life in mice was about five days for all linker-payload combinations; exposure was confirmed in non-human primates. A single-dose non-GLP study in cynomolgus monkeys showed only transient, mild changes in clinical chemistry.
Poster 5584 — TCX-101
TCX-101, a trispecific T-cell engager (TcE) co-targeting a tumor-associated carbohydrate (TACA) antigen and a tumor-associated protein antigen (TAA) for the treatment of solid tumors with enhanced preclinical activity.

- Four trispecific formats bound cancer cells more strongly than single-arm controls, which points to bivalent engagement of TACA and TAA.
- In co-culture with human T cells, the formats killed cancer cells of several origins in a concentration-dependent way. Knockout of either the TACA or the TAA reduced binding and killing.
