Six programmes, led by an ADC on its way to the clinic
TCX-201, an antibody-drug conjugate, is our first clinical candidate, with a clinical trial application planned for 2027. TCX-101 is a trispecific T-cell engager in preclinical development. Four further programmes target other TACAs.

The target glycan sits on proteins, is highly expressed in pancreatic, gastric and colorectal cancer and internalizes quickly, which suits payload delivery. In preclinical models the ADC reduced tumor growth dose-dependently in cell line- and patient-derived xenografts, including a metastatic colorectal cancer PDX, showed an antibody-like half-life of about five days and was well tolerated in a single-dose study in non-human primates. Selected in April 2026 as Tacalyx's first clinical candidate.

The TACA behind TCX-101 is anchored to sphingolipids and highly expressed in colon, lung, ovarian and breast cancer. It internalizes slowly, and an ADC format showed limited efficacy, so Tacalyx moved the antibody into T-cell engager formats: a 2+1 CD3 CrossMab in 2025, now a trispecific that binds the TACA and a tumor-associated protein antigen. In knockout cell lines, binding and T-cell-mediated killing depend on both targets.
